Ejaculated sperm are subjected to a variety of environmental stresses that can affect their reproductive capacity. They are also subject to changes in the microenvironment during storage and transit, potentially changing the content of sperm. MicroRNAs are a class of small non-coding RNAs that have the potential to modify the content and function of sperm. To determine the presence and diversity of microRNAs in mature human semen, we developed a microRNA library from ejaculated sperm obtained from healthy donors and identified a total of 138 microRNAs in the ejaculate. Most microRNAs were present at low levels, and most of the highly abundant sperm microRNAs represented all 18 of the conserved human microRNA families. There was significant overlap between ejaculated sperm and testis microRNAs, but only two microRNAs were present at higher levels in semen.
